Mitogenomes illuminate the origin and migration patterns of the indigenous people of the Canary Islands

The Canary Islands’ indigenous people have been the subject of substantial archaeological, anthropological, linguistic and genetic research pointing to a most probable North African Berber source. However, neither agreement about the exact point of origin nor a model for the indigenous colonization of the islands has been established. To shed light on these questions, we analyzed 48 ancient mitogenomes from 25 archaeological sites from the seven main islands. Most lineages observed in the ancient samples have a Mediterranean distribution, and belong to lineages associated with the Neolithic expansion in the Near East and Europe (T2c, J2a, X3a…). This phylogeographic analysis of Canarian ancient mitogenomes, the first of its kind, shows that some lineages are restricted to Central North Africa (H1cf, J2a2d and T2c1d3), while others have a wider distribution, including both West and Central North Africa, and, in some cases, Europe and the Near East (U6a1a1, U6a7a1, U6b, X3a, U6c1). In addition, we identify four new Canarian-specific lineages (H1e1a9, H4a1e, J2a2d1a and L3b1a12) whose coalescence dates correlate with the estimated time for the colonization of the islands (1st millennia CE). Additionally, we observe an asymmetrical distribution of mtDNA haplogroups in the ancient population, with certain haplogroups appearing more frequently in the islands closer to the continent. This reinforces results based on modern mtDNA and Y-chromosome data, and archaeological evidence suggesting the existence of two distinct migrations. Comparisons between insular populations show that some populations had high genetic diversity, while others were probably affected by genetic drift and/or bottlenecks. In spite of observing interinsular differences in the survival of indigenous lineages, modern populations, with the sole exception of La Gomera, are homogenous across the islands, supporting the theory of extensive human mobility after the European conquest

Abstract

One of the best documented Indo-European civilizations that inhabited Bulgaria is the Thracians, who lasted for more than five millennia and whose origin and relationships with other past and present-day populations are debated among researchers. Here we report 25 new complete mitochondrial genomes of ancient individuals coming from three necropolises located in different regions of Bulgaria – Shekerdja mogila, Gabrova mogila and Bereketska mogila – dated to II-III millennium BC. The identified mtDNA haplogroup composition reflects the mitochondrial variability of Western Eurasia. In particular, within the ancient Eurasian genetic landscape, Thracians locate in an intermediate position between Early Neolithic farmers and Late Neolithic-Bronze Age steppe pastoralists, supporting the scenario that the Balkan region has been a link between Eastern Europe and the Mediterranean since the prehistoric time. Spatial Principal Component Analysis (sPCA) performed on Thracian and modern mtDNA sequences, confirms the pattern highlighted on ancient populations, overall indicating that the maternal gene pool of Thracians reflects their central geographical position at the gateway of Europe.

В частности, в древнем евразийском генетическом ландшафте фракийцы занимают промежуточное положение между фермерами раннего неолита и степными скотоводами позднего неолита и бронзы, поддерживая сценарий, согласно которому Балканский регион является связующим звеном между Восточной Европой и Средиземноморьем с доисторического времени. Пространственный анализ основных компонентов (sPCA), проведенный на фракийских и современных последовательностях мтДНК, подтверждает паттерн, выделенный на древних популяциях, что в целом указывает на то, что материнский генофонд фракийцев отражает их центральное географическое положение у ворот Европы.

Abstract

Objectives

In the 14th century AD, medieval Europe was severely affected by the Great European Famine as well as repeated bouts of disease, including the Black Death, causing major demographic shifts. This high volatility led to increased mobility and migration due to new labor and economic opportunities, as evidenced by documentary and stable isotope data. This study uses ancient DNA (aDNA) isolated from skeletal remains to examine whether evidence for large‐scale population movement can be gleaned from the complete mitochondrial genomes of 264 medieval individuals from England (London) and Denmark.

Materials and Methods

Using a novel library‐conserving approach to targeted capture, we recovered 264 full mitochondrial genomes from the petrous portion of the temporal bones and teeth and compared genetic diversity across the medieval period within and between English (London) and Danish populations and with contemporary populations through population pairwise ΦST analysis.

Results

We find no evidence of significant differences in genetic diversity spatially or temporally in our dataset, yet there is a high degree of haplotype diversity in our medieval samples with little exact sequence sharing.

Discussion

The mitochondrial genomes of both medieval Londoners and medieval Danes suggest high mitochondrial diversity before, during and after the Black Death. While our mitochondrial genomic data lack geographically correlated signals, these data could be the result of high, continual female migration before and after the Black Death or may simply indicate a large female effective population size unaffected by the upheaval of the medieval period. Either scenario suggests a genetic resiliency in areas of northwestern medieval Europe.

Митохондриальные геномы как средневековых лондонцев, так и средневековых датчан свидетельствуют о высоком митохондриальном разнообразии до, во время и после Черной Смерти. Goth migration induced changes in the matrilineal genetic structure of the central-east European population

Abstract

For years, the issues related to the origin of the Goths and their early migrations in the Iron Age have been a matter of hot debate among archaeologists. Unfortunately, the lack of new independent data has precluded the evaluation of the existing hypothesis. To overcome this problem, we initiated systematic studies of the populations inhabiting the contemporary territory of Poland during the Iron Age. Here, we present an analysis of mitochondrial DNA isolated from 27 individuals (collectively called the Mas-VBIA group) excavated from an Iron Age cemetery (dated to the 2nd-4th century A.D.) attributed to Goths and located near Masłomęcz, eastern Poland. We found that Mas-VBIA has similar genetic diversity to present-day Asian populations and higher diversity than that of contemporary Europeans. Our studies revealed close genetic links between the Mas-VBIA and two other Iron Age populations from the Jutland peninsula and from Kowalewko, located in western Poland. We disclosed the genetic connection between the Mas-VBIA and ancient Pontic-Caspian steppe groups. Similar connections were absent in the chronologically earlier Kowalewko and Jutland peninsula populations. The collected results seem to be consistent with the historical narrative that assumed that the Goths originated in southern Scandinavia; then, at least part of the Goth population moved south through the territory of contemporary Poland towards the Black Sea region, where they mixed with local populations and formed the Chernyakhov culture. Finally, a fraction of the Chernyakhov population returned to the southeast region of present-day Poland and established the archaeological formation called the “Masłomęcz group”.

Самый интересный момент:

Интересно отметить, что частоты гаплогрупп мтДНК U5a (характерные для групп восточных охотников-собирателей) и U5b (характерные для групп западных охотников-собирателей) значительно отличались в Mas-VBIA и других популяциях IA, изученных до настоящего времени (JIA и Kow-OVIA). ). Mas-VBIA характеризовался отсутствием гаплогруппы мтДНК U5b и высокой частотой гаплогруппы мтДНК U5a (14,8%), тогда как в JIA и Kow-OVIA гаплогруппа мтДНК U5b была более распространенной, чем U5a. Как отмечалось ранее, доминирование U5b над U5a в Центральной Европе наблюдалось только во время IA. Следовательно, либо процесс, который привел к временному увеличению частоты гаплогруппы мтДНК U5b, не затронул регионы к востоку от Вислы, либо его последствия были впоследствии устранены другим демографическим событием.

""Масломенчский" - представлен исключительно в женских сериях преимущественно Западной Украины и Молдавии. Несколько уклоняется от западных типов за счёт слегка подуплощённого лица. Истоки его можно видеть только на нижней Висле в вельбаркских могильниках. В славянскую эпоху данный тип получает широкое распространение, начинает появляться в мужских сериях и становится обычным среди женских славянских групп на Западной Украине и в смежных районах Польши. Среди мужчин славянской эпохи он тоже встречается, но реже, чем у женщин.
Судя по всему это какое-то местное доготское население захваченное миграцией и в дальнейшем послужившее субстратом для славянского (в женских славянских сериях он представлен сильнее, чем в мужских). Особо интересна его некоторая "лапоноидность" выражающаяся в снижении горизонтальной профилировки и увеличении ширины лица.


Существенно отличаются между собой по составу мужская и женская серии Масломенческой группы. Мужская серия состоит, главным образом, из носителей средиземноморского антропологического типа, а в женской серии доминирующим является лапоноидный тип (W. Kozak-Zychman).Интересно, что в средневековых сериях с территории Любельщины (Грудек Надбужный) лапоноидный тип доминирует как в мужской, так и в женской сериях. Это дает основание утверждать, что по крайней мере женская часть населения, похороненного на могильниках вельбарськой культуры Грудека и Масломенча, может быть генетически связана с населением этого региона эпохи средневековья"."

Abstract

Human ancient DNA studies have revealed high mobility in Europe’s past, and have helped to decode the human history on the Eurasian continent. Northeastern Europe, especially north of the Baltic Sea, however, remains less well understood largely due to the lack of preserved human remains. Finland, with a divergent population history from most of Europe, offers a unique perspective to hunter-gatherer way of life, but thus far genetic information on prehistoric human groups in Finland is nearly absent. Here we report 103 complete ancient mitochondrial genomes from human remains dated to AD 300–1800, and explore mtDNA diversity associated with hunter-gatherers and Neolithic farmers. The results indicate largely unadmixed mtDNA pools of differing ancestries from Iron-Age on, suggesting a rather late genetic shift from hunter-gatherers towards farmers in North-East Europe. Furthermore, the data suggest eastern introduction of farmer-related haplogroups into Finland, contradicting contemporary genetic patterns in Finns.

Abstract

Two ancient Egyptian child mummies at the University of Tartu Art Museum (Estonia) were, according to museum records, brought to Estonia by the young Baltic-German scholar Otto Friedrich von Richter, who had travelled in Egypt during the early 19th century. Although some studies of the mummies were conducted, a thorough investigation has never been made. Thus, an interdisciplinary team of experts studied the remains using the most recent analytical methods in order to provide an exhaustive analysis of the remains. The bodies were submitted for osteological and archaeothanatological study, radiological investigation, AMS radiocarbon dating, chemical and textile analyses, 3D modelling, entomological as well as aDNA investigation. Here we synthesize the results of one of the most extensive multidisciplinary analyses of ancient Egyptian child mummies, adding significantly to our knowledge of such examples of ancient funerary practices.

Две древнеегипетские детские мумии из художественного музея Тартуского университета (Эстония) были, согласно музейным записям, привезены в Эстонию молодым прибалтийско-немецким ученым Отто Фридрихом фон Рихтером, который путешествовал по Египту в начале XIX века. Abstract

The clarification of the genetic origins of present-day Tibetans requires an understanding of their past relationships with the ancient populations of the Tibetan Plateau. Here we successfully sequenced 67 complete mitochondrial DNA genomes of 5200 to 300-year-old humans from the plateau. Apart from identifying two ancient plateau lineages (haplogroups D4j1b and M9a1a1c1b1a) that suggest some ancestors of Tibetans came from low-altitude areas 4750 to 2775 years ago and that some were involved in an expansion of people moving between high-altitude areas 2125 to 1100 years ago, we found limited evidence of recent matrilineal continuity on the plateau. Furthermore, deep learning of the ancient data incorporated into simulation models with an accuracy of 97% supports that present-day Tibetan matrilineal ancestry received partial contribution rather than complete continuity from the plateau populations of the last 5200 years.

Abstract

Objective

In this work we aim to investigate the origins and genetic affinities of Bronze Age populations (2,400–1,100 BC) from the region of southern Poland and to trace maternal kinship patterns present in the burials of those populations by the use of complete mitochondrial genomes.

Materials and methods

We performed ancient DNA analyses for Bronze Age individuals from present‐day Poland associated with the Strzyżow culture, the Mierzanowice culture, and the Trzciniec Cultural circle. To obtain complete mitochondrial genomes, we sequenced genomic libraries using Illumina platform. Additionally, hybridization capture was used to enrich some of the samples for mitochondrial DNA. AMS 14C‐dating was conducted for 51 individuals to verify chronological and cultural attribution of the analyzed samples.

Results

Complete ancient mitochondrial genomes were generated for 80 of the Bronze Age individuals from present‐day Poland. The results of the population genetic analyses indicate close maternal genetic affinity between Mierzanowice, Trzciniec, and Corded Ware culture‐associated populations. This is in contrast to the genetically more distant Strzyżów people that displayed closer maternal genetic relation to steppe populations associated with the preceding Yamnaya culture and Catacomb culture, and with later Scythians. Potential maternal kinship relations were identified in burials of Mierzanowice and Trzciniec populations analyzed in this study.

Discussion

Results revealed genetic continuity from the Late Neolithic Corded Ware groups to Bronze Age Mierzanowice and Trzciniec‐associated populations, and possible additional genetic contribution from the steppe to the formation of the Strzyżów‐associated group at the end of 3rd millennium BC. Mitochondrial patterns indicated several pairs of potentially maternally related individuals mostly in Trzciniec‐associated group.

Аннотация

На фоне активного изучения генетического состава ранних кочевников с территории Алтае-Саянской горной системы ряд популяций скифского времени из прилегающей лесостепной зоны остаются не исследованными методами палеогенетики. В статье представлены первые результаты палеогенетического анализа носителей староалейской культуры из могильника Фирсово-XIV в Барнаульском Приобье. Рассмотрение небольшой серии образцов митохондриальной ДНК (N=10) позволил подтвердить участие популяций, связанных с автохтонным генетическим субстратом южных районов Западной Сибири, в формировании генетического состава староалейского населения (специфичный состав западно-евразийского компонента генофонда мтДНК и присутствие автохтонной А10 гаплогруппы). Установлено присутствие в староалейской популяции вариантов мтДНК (линии гаплогрупп A8 и A11), которые свидетельствуют о ее связях с носителями скифо-сибирского круга культур, населявших в раннем железном веке территории к востоку от Верхнего Приобья – Алтае-Саянскую горную систему, Туву и прилегающие районы Центральной Азии. Палеогенетические данные свидетельствуют, что генетический состав носителей староалейской популяции формировался в условиях взаимодействия автохтонных популяций региона, генетические корни которых уходят в эпоху бронзы, и пришлых групп, являвшихся носителями культурных традиций ранних кочевников скифского времени. Учитывая информативность первых палеогенетических результатов, можно ожидать существенной детализации реконструкций этногенетических процессов по мере увеличения численности образцов ДНК из староалейской популяции, анализа дополнительных генетических маркеров (Y-хромосомы) и получения данных о генофонде других социумов, населявших Верхнее Приобье и сопредельные регионы Южной Сибири в раннем железном веке.

Abstract

Xinjiang is a key region in northwestern China, connecting East and West Eurasian populations and cultures for thousands of years. To understand the genetic history of Xinjiang, we sequenced 237 complete ancient human mitochondrial genomes from the Bronze Age through Historical Era (41 archaeological sites). Overall, the Bronze Age Xinjiang populations show high diversity and regional genetic affinities with Steppe and northeastern Asian populations along with a deep ancient Siberian connection for the Tarim Basin Xiaohe individuals. In the Iron Age, in general, Steppe-related and northeastern Asian admixture intensified, with North and East Xinjiang populations showing more affinity with northeastern Asians and South Xinjiang populations showing more affinity with Central Asians. The genetic structure observed in the Historical Era of Xinjiang is similar to that in the Iron Age, demonstrating genetic continuity since the Iron Age with some additional genetic admixture with populations surrounding the Xinjiang region.

Maternal genetic origin of the late and final Neolithic human populations from present-day Poland

Objective
We aim to identify maternal genetic affinities between the Middle to Final Neolithic (3850–2300 BC) populations from present-day Poland and possible genetic influences from the Pontic steppe.

Materials and methods
We conducted ancient DNA studies from populations associated with Złota, Globular Amphora, Funnel Beaker, and Corded Ware cultures (CWC). We sequenced genomic libraries on Illumina platform to generate 86 complete ancient mitochondrial genomes. Some of the samples were enriched for mitochondrial DNA using hybridization capture.

Results
The maternal genetic composition found in Złota-associated individuals resembled that found in people associated with the Globular Amphora culture which indicates that both groups likely originated from the same maternal genetic background. Further, these two groups were closely related to the Funnel Beaker culture-associated population. None of these groups shared a close affinity to CWC-associated people. Haplogroup U4 was present only in the CWC group and absent in Złota group, Globular Amphora, and Funnel Beaker cultures.

Discussion
The prevalence of mitochondrial haplogroups of Neolithic farmer origin identified in Early, Middle and Late Neolithic populations suggests a genetic continuity of these maternal lineages in the studied area. Although overlapping in time – and to some extent – in cultural expressions, none of the studied groups (Złota, Globular Amphora, Funnel Beaker), shared a close genetic affinity to CWC-associated people, indicating a larger extent of cultural influence from the Pontic steppe than genetic exchange. The higher frequency of haplogroup U5b found in populations associated with Funnel Beaker, Globular Amphora, and Złota cultures suggest a gradual maternal genetic influx from Mesolithic hunter-gatherers. Moreover, presence of haplogroup U4 in Corded Ware groups is most likely associated with the migrations from the Pontic steppe at the end of the Neolithic and supports the observed genetic distances.
