Аполипопротеин E в популяциях мира

Как и обещал, вот пример того, что можно сделать данными из базы AlFreD (Allele Frequency Database).
Одними из самых наиболее изученных  (по количеству исследованных популяций) аллелей являются аллели локуса гена апоЕ человека, который локализуется в хромосоме 19 и находится в кластере с другими аполипопротеинами апоС1 и апоС2.
Ген состоит из 4 экзонов, 3 интронов, 3597 пар нуклеотидов. Ген характеризуется полиморфизмом. Существуют 3 основных аллеля апоЕ: нормальная AПОE-?3, AПОE-?2, не способная связываться с ЛПНП-рецептором и AПОE-?4. Аллели отличаются друг от друга лишь точечной мутацией. AПОE-?2 связан с гиперлипопротеинемией III типа и повышенным риском атеросклероза. AПОE-?4 связан с повышенным уровнем холестерина в крови и задержкой в регенерации нейронов, является главным генетическим риском болезни Альцгеймера.

Ген AПОE регулирует синтез елок апоЕ человека состоит из 299 аминокислот, молекулярная масса — 34 кДа. Состоит из двух доменов: один связывается с липидом, а второй определяет связывание с апоЕ-рецепторами и клеточным гликокаликсом. Синтезируется в основном в печени и мозге, причём эти два пула апоЕ совершенно не зависимы друг от друга, так как апоЕ не способен преодолеть барьер между кровью и церебро-спинальной жидкостью. В крови апоЕ определяет поглощение ремнантов хиломикрон и ремнантов ЛПОНП печенью. В мозге апоЕ синтезируется астроцитами и микроглией, а рецепторы к нему экспрессируются нейронами. Таким образом апоЕ осуществляет доставку холестерина к месту миелинизации. В настоящий момент у млекопитающих идентифицировано семь рецепторов, лигандом которых является АпоЕ. Все эти рецепторы принадлежат к эволюционно-консервативному семейству рецепторов липопротеинов низкой плотности.

Аллель AПОE-?4 значительно повышает риск развития болезни Альцгеймера. От 40 до 65 % больных имеют по крайней мере одну копию апоЕ4, тогда как только 25 % всей популяции имеют этот аллель. Неизвестно, как апоЕ4 повышает риск развития болезни Альгеймера.

В ходе крупного генетического исследования было обнаружено, что у носителей аллели AПОE-?4 на риск заболевания влияют некоторые вариации гена GAB2.

Ген аполипопротеина Е (APOE) участвует в регуляции уровня холестерина крови.
Один из его аллелей, называемый APOE*e4, ассоциирован с повышенным уровнем холестерина и рассматривается как «экономичный» (Mahley, Rall 2000; Corbo, Scacchi 1999). Этот аллель APOE*e4 является фактором риска болезни Альцгеймера и сердечно-сосудистых заболеваний у жителей Европы и евроамериканцев. Однако у племен суб-Сахарской Африки этот аллель не является фактором риска и не ассоциирован с высоким уровнем холестерина. У афроамериканцев ассоциация наблюдается, однако более слабая, чем у евроамериканцев (см. обзор в: Corbo, Scacchi, 1999).
 Это свидетельствует о разном проявлении одного и того же аллеля в различных условиях среды и на различном генетическом фоне.
Частоты аллеля APOE*e4 варьируют от 5–15 % у большинства европейских народов до 40 % – у автохтонных народов Африки, Австралии и Америки (Gerders et al. 1996; Corbo, Scacchi, 1999, Jaramillo-Correa et al. 2001). Можно полагать, что APOE*e4, являющийся более древним, предковым аллелем, был вытеснен в популяциях с производящим хозяйством более молодыми производными от него аллелями APOE*e3 (ассоциированным со средним уровнем холестерина) и APOE*e2 (ассоциированным с низким уровнем холестерина).
При этом маловероятно, что болезнь Альцгеймера или сердечно-сосудистые заболевания были факторами отбора, так как эти заболевания проявляются в пострепро-дуктивный период (Corbo, Scacchi 1999). Вероятно, какой-либо другой признак, ассоциированный с этими аллелями и повышающий выживаемость в дорепродуктивный период, опосредовал действие естественного отбора.

Статистика корреляции Пирсона между частотами трех типов аллелей АПОЕ.
В ходе статистического анализа по полученным значениям коэффициента детерминации
и шклаы Чеддока, опеределенно наличие умеренной корреляции (0.36) между частотами распространения APOE*2 (Var1) и APOE*3(Var2), а также близкой к значимой корреляции (0.59) между частотами распространения APOE*3(Var2) и APOE*4(Var3).
Нужно однако помнить о том, что при значениях показателей тесноты связи меньше 0,7 величина коэффициента детерминации всегда будет ниже 50 %. Это означает, что на долю вариации факторных признаков приходится меньшая часть по сравнению с остальными неучтенными в модели факторами, влияющими на изменение результативного показателя. Построенные при таких условиях регрессионные модели имеют низкое практическое значение.
